Test Description
Genomic Unity® Precision Anesthesia Analysis is a targeted test that uses a whole genome platform to detect all major clinically relevant variant types from a single sample. This targeted test focuses on 22 genes associated with possible anesthetic management implications, enabling providers to implement targeted and preventive pathways.
Genomic Unity® Precision Anesthesia Analysis provides a single, unified clinical report that replaces a battery of tests including: targeted gene panel, single gene analysis and multiplex ligation dependent probe amplification (MLPA), as well as PCR and southern blot tests for selected short tandem repeat expansions and analysis of the MT-ND4 gene m.11232T>C; p.L158P variant, including heteroplasmy levels.
When to Order
Order this test for patients planning to undergo general anesthesia or sedation with a personal or family history of life-threatening anesthetic crises or complications, unexplained sudden cardiac death, or maternal Venezuelan lineage. Key indications include malignant hyperthermia episodes, prolonged neuromuscular paralysis, adverse neurological reactions, unprovoked syncope, or arrhythmias.
Included Genes
BCHE, CACNA1S, CALM1, CALM2, CALM3, CASQ2, CNBP, DSC2, DSG2, DMPK, DSP, KCNH2, KCNQ1, PHOX2B, PKP2, PLN, RYR1, RYR2, SCN5A, STAC3, TMEM43, TRDN
Mitochondrial genes
MT-ND4 gene m.11232T>C; p.L158P variant.

Methods and Limitations
Genomic Unity® Precision Anesthesia Analysis uses a PCR-free whole genome sequencing (WGS) platform paired with our Genomic Intelligence® analytical software.
SNVs:
>99.9% sensitivity
>99.9% specificity
>99.8% positive predictive value
Structural variants:
96% clinical sensitivity
Test limitations:
The following gene is not fully covered: KCNQ1. Therefore there may be sequence changes or structural variants in certain regions of this gene that are not identified with this test.
